چکیده

Escherichia coli O157:H7, an emerging cause of food-borne disease, has now been reported from several countries worlwide. Cattle and sheep are known as a principle reservoir of this agent. It is also prevalent in gastrointestinal tract of other animals like pigs, dogs, cats and birds. This agent can be transmitted to humans by food and water and even person to person contact. The random amplified polymorphic DNA (RAPD) technique has been applied to evaluate zoonotic potency of Escherichia coli O157:H7 local isolates. Ten random decamer primers have been evaluated in this study to differentiate isolates originated from humans and animals. The OPA-02, OP-03, OPA-04, OPA07, OPA-08, OPA-09, OPA-10, OPA-13, OPA-19, and OPA-20 decamer primers show a good differentiation pattern of E. coli O157:H7 isolated from human, cattle and chicken feces, as well as from beef. The primers constantly produce 16, 16, 11, 14, 8, 24, 7, 11, 7, and 12 polymorphic bands, respectively. Various human isolates that were isolated from clinically ill and healthy patients share common genetic clusters with animal isolates from beef, as well as cattle and chicken feces with genetic similarity coefficients greater than 70%. According to this result, It has been concluded the transmission of E. coli O157:H7 local isolates from animals to humans is potential occur.

Differentiation of avian pathogenic Escherichia coli (APEC) strains by random amplified polymorphic DNA (RAPD) analysis.

Here we describe the application of a random amplified polymorphic DNA (RAPD) analysis for molecular genetic typing avian pathogenic Escherichia coli (APEC) strains. The RAPD technique was shown to be highly reproducible. Stable banding patterns with a high discriminatory capacity were obtained using two different primers. Overall, 55 E. coli strains were analyzed with a RAPD technique. The RAP...

Isolation and characterization Shiga toxin-producing Escherichia coli from sheep and goats in Jordan with evidence of multiresistant serotype O157:H7

Ninety-three rectal swabs of lambs and young goats from two extensively and two intensively managed herds in Jordan were taken and examined for Shiga toxin-producing Escherichia coli (STEC). The bacteriological examination included the preenrichment of rectal swabs in EC broth with novobiocin, and a subsequent parallel isolation on enterohemolysin agar and immunomagnetic separation with cultiva...

Polymorphic amplified typing sequences provide a novel approach to Escherichia coli O157:H7 strain typing.

Escherichia coli O157:H7 (O157) strains are commonly typed by pulsed-field gel electrophoresis (PFGE) following digestion of genomic DNA with the restriction enzyme XbaI. We have shown that O157 strains differ from each other by a series of discrete insertions or deletions, some of which contain recognition sites for XbaI, suggesting that these insertions and deletions are responsible for the d...

Clonal nature of enterotoxigenic Escherichia coli serotype O6:H16 revealed by randomly amplified polymorphic DNA analysis.

The genetic relatedness among 29 enterotoxigenic Escherichia coli strains of serotype O6:H16 was investigated by randomly amplified polymorphic DNA (RAPD) analysis. The strains were isolated in different parts of the world, displayed CS1-CS3 or CS2-CS3 profiles, and expressed heat-labile toxin (LT) and heat-stable toxin; a single strain expressed only LT. Ten RAPD types were distinguished and s...

Genetic diversity among Elaeagnus angustifolia L. populations based on some morphological traits and Random Amplified Polymorphic DNA Markers

Elaeagnus angustifolia L. is a Eurasian tree that has become naturalized and has various ecological, medicinal and economical uses. In this study, a combination of morphological traits and RAPD markers were used to study the presence or absence of an association between genetic variation and morphological features among five populations of E. angustifolia collected from the East Azarbaijan of I...
